Skin Infection Model

A skin infection model is an experimental system that reproduces key features of microbial invasion and host defense in skin, helping researchers study infection in a controlled setting. Depending on its design, the model combines cultured keratinocytes, reconstructed skin tissue, or animal skin with a selected pathogen; infection can then be assessed through barrier disruption, microbial growth, inflammatory signaling, and immune-cell recruitment. In immunology and infection research, these models clarify how epithelial barriers and innate immune responses limit disease while enabling evaluation of antimicrobial treatments, virulence mechanisms, and therapeutic strategies before clinical testing. Their controlled conditions also support reproducible comparisons across pathogens and interventions.

Generating an Ex Vivo Ovine Wounded Skin Model Infected with a Bacterial Pathogen

2025

The video demonstrates the generation of an ex vivo ovine skin model infected with Staphylococcus aureus. This entails treating a lamb's forehead skin biopsy section with antibiotics, establishing a wound bed, positioning it on a permeable membrane in a multiwell plate with antibiotic-free medium, and establishing bacterial infection in the skin wound.

Developing a Mouse Model of Chronic Pseudomonas aeruginosa Skin Wound Infection

Source: de Vries, C. R., et.al. A Delayed Inoculation Model of Chronic Pseudomonas aeruginosa Wound Infection. J. Vis. Exp. (2020)This video demonstrates the delayed inoculation of luminescent Pseudomonas aeruginosa into a mouse bearing two full-thickness dorsal skin wounds covered by a transparent film dressing. This facilitates biofilm formation and the establishment of chronic infection. Regular bioluminescence monitoring enables quantification of bacterial burden and tracking of infection...

Cultivation of Pathogenic Spirochetes to Investigate Skin Infection

Exp.(2022)This video demonstrates the cultivation of pathogenic spirochetes from infected skin tissue to model dermal infection. It highlights their localization in the dermis, interaction with the extracellular matrix, and survival during selective culture conditions.

A Murine Skin Wound Infection Model to Study the Immune Response against a Bacterial Pathogen

This video demonstrates a technique to generate a murine model of skin wound infection. Upon infecting mice — engineered to express a fluorescent protein in neutrophils — with a genetically modified bioluminescent Staphylococcus aureus — a pathogenic bacteria. The bacterial burden at the infection site and the neutrophil recruitment dynamics can be studied using whole animal imaging.

Staphylococcal Skin Infections

Staphylococcus aureus is a Gram-positive coccus that resides harmlessly on the skin and mucous membranes of healthy individuals. When the skin barrier is breached, it can shift from a commensal to an opportunistic pathogen. This transition is facilitated by surface adhesins, such as clumping factor B and S. aureus surface protein G (SasG), which bind to structural proteins, including loricrin and cytokeratin, in the damaged epidermis. Protein A, another key factor, binds the Fc region of...
