Checkerboard Assay

The checkerboard assay is a laboratory method used to assess whether two antimicrobial agents interact synergistically, additively, or antagonistically when used together. It tests serial concentration combinations of the agents in a two-dimensional matrix, typically using microbial growth inhibition to determine the minimum inhibitory concentration for each combination and calculate a fractional inhibitory concentration index. In immunology and infection research, this approach helps characterize combination therapies, identify dose-sparing interactions, and evaluate strategies for overcoming antimicrobial resistance. Results can guide experimental treatment design and support the development of more effective approaches to controlling infectious organisms.

The measurement protocol and data analysis procedure are given for obtaining transverse coherence of a synchrotron radiation X-ray source along four directions simultaneously using a single 2-D checkerboard phase grating. This simple technique can be applied for complete transverse coherence characterization of X-ray sources and X-ray optics.

We developed the Visual-Motor Response to quantitate the motor output of larval zebrafish in response to light increments and decrements. We also examined zebrafish vision mutants, including the no optokinetic response (nrc) mutants, which were thought to be completely blind when tested by another vision assay, the optokinetic reflex.

Antimicrobial synergy testing is used to evaluate the effect of two or more antibiotics used in combination and is typically performed by one of two methods: the checkerboard array or the time-kill assay. Here, we present an automated, inkjet printer-assisted checkerboard array synergy technique and a classic time-kill synergy study.
