Sporozoite Harvest

Sporozoite harvest is the process of collecting infectious sporozoites, the motile stage of malaria parasites that develops in mosquito salivary glands, for immunology and infection research. In a typical workflow, infected mosquitoes are dissected, salivary glands are removed, and gentle mechanical disruption releases sporozoites into a collection medium, followed by clarification or purification when needed. Researchers use harvested parasites to study host invasion, parasite development, vaccine-induced immunity, and antibody or cellular responses. Consistent recovery and careful handling help preserve sporozoite viability, supporting reproducible infection models and evaluation of interventions that target transmission or early liver-stage infection.

This video demonstrates the generation of an experimental mouse model of cerebral malaria. Sporozoites isolated from Plasmodium berghei-infected female mosquitoes are injected into a restrained mouse's tail veins, leading to blood-brain barrier disruption and brain inflammation, causing cerebral malaria. This infected mouse model offers a valuable tool to study in vivo microvascular pathology associated with cerebral malaria.

We describe here a protocol for isolating myeloid cells from mouse skin and draining lymph node following intradermal injection of Plasmodium sporozoites. Flow cytometry of collected cells provides a reliable assay to characterize the skin and draining lymph node inflammatory response to the parasite.
