Bone Graft Materials

Bone graft materials are biological or synthetic substances used to repair bone defects and support new tissue formation when bone cannot heal or regenerate adequately on its own. They work by providing a three-dimensional scaffold for cell attachment and vascular ingrowth, while some materials also supply osteogenic cells or signals that stimulate osteogenesis; successful integration depends on material properties, defect environment, and host response. In medicine, autografts, allografts, xenografts, and synthetic substitutes are selected for fracture repair, spinal fusion, and dental or craniofacial reconstruction. Their development aims to balance structural strength, biocompatibility, resorption, and predictable bone replacement.

Bone metastasis models do not develop metastasis uniformly or with a 100% incidence. Direct intra-osseous tumor cell injection can result in embolization of the lung. We present our technique modeling primary bone tumors and bone metastasis using solid tumor graft implantation into bone, leading to reproducible engraftment and growth.

This video demonstrates the induction of intestinal graft-versus-host disease, GvHD, in a lethally-irradiated recipient mouse using allogeneic hematopoietic stem cells and GvHD-inducing splenic alloreactive T lymphocytes. This protocol helps establish a severe acute intestinal GvHD model to understand the mechanism of the disease and develop innovative therapeutic options.

Here we present a surgical protocol in rabbits with the aim to assess bone substitution materials in terms of bone regeneration capacities. By using PEEK cylinders fixed onto rabbit skulls, osteoconduction, osteoinduction, osteogenesis and vasculogenesis induced by the materials may be evaluated either on live or euthanized animals.

In this video, we describe a method for the detection of osteoblast-secreted calcium deposits using an anionic dye, Alizarin Red S. The cultured osteoblasts are first stained and then the dye is recovered for colorimetric quantification of calcium deposits or the degree of osteoblast mineralization.
