Congo Red Staining

Congo red staining is a histopathological technique used to detect amyloid deposits, abnormal accumulations of protein that can damage tissues and organs. The dye binds to the ordered β-pleated sheet structure of amyloid fibrils, producing characteristic salmon-pink to red deposits in routine microscopy and apple-green birefringence when viewed under polarized light. In medicine, this method supports the diagnosis of systemic and localized amyloidosis in tissue biopsies, helps distinguish amyloid from other extracellular materials, and provides a foundation for assessing disease distribution and treatment response. Its interpretation is strengthened by correlating staining patterns with clinical and laboratory findings.
