Langendorff Apparatus

The Langendorff apparatus is an ex vivo perfusion system that keeps an isolated heart functioning by supplying an oxygenated physiological solution, making it valuable for studying cardiac performance without whole-animal influences. Perfusate enters the aorta in a retrograde direction, closes the aortic valve, and flows through the coronary arteries to deliver oxygen and nutrients to the myocardium; pressure or flow can be controlled while ventricular function is recorded. In medicine and cardiovascular research, this preparation supports investigations of ischemia-reperfusion injury, myocardial metabolism, drug effects, and recovery after cardiac stress, providing a controlled platform for linking coronary perfusion with heart function.

In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.

Operant drug self-administration and conditioned place preference (CPP) procedures are expansively used in research to model various components of drug reinforcement, consumption, and addiction in humans. In this report, we combined traditional CPP and self-administration methods as a novel approach to studying drug reinforcement and addiction in rats.
