Site-specific Tumor Inoculation

Site-specific tumor inoculation is a research technique that introduces tumor cells or tissue into a defined anatomical location to model cancer growth in its native or clinically relevant context. By placing cells at a selected site, the method preserves interactions with local stroma, vasculature, immune cells, and extracellular matrix, which can influence tumor establishment and progression. This approach supports studies of tumor biology, invasion, metastasis, treatment response, and surgical or imaging strategies in preclinical medicine. Compared with implantation at non-native sites, site-specific models can improve biological relevance while helping researchers evaluate how anatomical context shapes disease and therapeutic outcomes.

An In Vitro Assay to Evaluate the Anti-Tumor Activity of Tumor-Specific CAR T Cells

This video demonstrates an in vitro assay to validate chimeric antigen receptor (CAR) T cell activity. B-cell lymphoma cells expressing CD19 antigen and cytoplasmic luciferase are co-cultured with CD19-specific CAR T cells. CAR binding to CD19 on a tumor induces the CAR T cells to release cytokines and toxins that cause tumor cell cytotoxicity. The CAR T cell activity is evaluated by quantifying the released cytokine and decreased luciferase-mediated bioluminescence in the tumor cells.

To clarify the multistep process of peritoneal dissemination of ovarian carcinoma, the current study presents a murine experimental model of original tumor development and peritoneal metastasis via orthotopic inoculation with these tumor cells.

This video demonstrates a technique to study the in vivo localization of tumor-specific antibodies in a mouse tumor xenograft model. Upon subcutaneously injecting cancer cells into mice for tumor formation, fluorescently-labeled antibodies are injected that bind to tumor-specific receptors, enabling in vivo assessment of antibody localization via imaging.
