Transthoracic Injection

Transthoracic injection is a medical technique that delivers a drug, biologic, contrast agent, or experimental material through the chest wall into a selected thoracic structure or space. The procedure uses a needle advanced across the skin and intercostal tissues toward the target, with anatomical landmarks or imaging guidance helping control placement and injection depth. In medicine and biomedical research, transthoracic injection supports localized treatment, delivery to organs such as the heart or lungs, and experimental modeling of disease or tissue repair. Accurate targeting can improve local exposure while limiting systemic distribution, although careful planning is essential to reduce risks such as bleeding, pneumothorax, or injury to adjacent structures.

Transthoracic echocardiography offers a noninvasive method for the evaluation of cardiac function in mice. A combination of ultrasound and Doppler imaging modalities can be used to obtain dimensional measurements of the heart and intracardiac blood flow, which together provide an assessment of cardiac systolic and diastolic performance.

Speckle tracking echocardiography is an emerging diagnostic imaging technique for the quantitative assessment of global and regional myocardial performance. Standard view echocardiographic motion images are recorded and deformation parameters are subsequently measured by automated continuous frame-by-frame tracking and motion analysis of speckles within the B-mode images of the myocardium.

Here we describe, step by step, a detailed protocol for performing echocardiography in the rabbit model. We show how to correctly obtain the different echocardiographic views and imaging planes, as well as the different imaging modes available in a clinical echocardiography system routinely used in human and veterinary patients.
