Bbb Mini Brain

A BBB mini brain is a three-dimensional in vitro neuroscience model that combines brain-like neural tissue with blood-brain barrier components to reproduce key features of the human brain’s protective interface. In this model, brain endothelial cells form tight junctions that regulate molecular transport, while neural and supporting cells influence barrier formation, permeability, and tissue function. BBB mini brains provide a controlled platform for studying barrier development, brain disease, neurotoxicity, and drug delivery. By reducing reliance on animal models and better representing human cellular interactions, they can improve the evaluation of therapeutics designed to reach the central nervous system.

The aim of the present study was to validate the reproducibility of an in vitro BBB model involving a rat syngeneic co-culture of endothelial cells and astrocytes. The endothelial cell monolayer presented high TEER and low LY permeability. Expression of specific TJ proteins, functional responses to inflammation and functionality of transporters and receptors were assessed.

A common protein extraction protocol using urea/thiourea/SDS buffer for human and mice brain tissue allows indentification of proteins by 2D-DIGE and their subsequent characterization by mini 2DE immunoblotting. This method enables one to obtain more reproducible and reliable results from human biopsies and experimental models.
