Brain Tissue Analysis

Brain tissue analysis is the systematic examination of brain structure, cellular composition, molecular features, and pathology to understand how nervous tissue functions and changes. Researchers preserve and section tissue, apply histological stains, antibodies, or molecular assays, and use microscopy or image analysis to identify neurons, glial cells, proteins, lesions, and patterns of gene expression. In neuroscience, these approaches connect anatomy with neural activity, development, disease, and injury, supporting research on neurodegenerative disorders, tumors, and traumatic damage while helping evaluate experimental treatments. Careful sampling, controls, and quantitative analysis improve the reliability and biological interpretation of findings.

Source: Wulf, M., et.al. Laser Microdissection-Based Protocol for the LC-MS/MS Analysis of the Proteomic Profile of Neuromelanin Granules. J. Vis. Exp. (2021).This video demonstrates the analysis of peptide samples from neuromelanin granules (NMGs) isolated from human postmortem brain tissue. The peptides are first separated using high-performance liquid chromatography (HPLC) and then analyzed by mass spectrometry to determine their mass-to-charge ratios (m/z). Targeted peptides are fragmented...

Until recently, expression studies on human brain were limited to quantification of RNA or protein. With the chromatin immunoprecipitation techniques described in this paper, it will be possible to map histone methylation and other epigenetic regulators of gene expression in postmortem brain.

A Hydrophobic Tissue Clearing Method for a Rat Brain Tissue Sample

Source: Kirchner, K.N., et al., A Hydrophobic Tissue Clearing Method for Rat Brain Tissue. J. Vis. Exp. (2020).This video demonstrates the method of hydrophobic tissue clearing of a rat brain tissue sample. The sample is sequentially dehydrated in methanol, treated with a dichloromethane and methanol mixture to dissolve lipids and pigments, and transferred to dibenzyl ether for complete tissue clearing, enabling detailed imaging without thin sectioning.
