Matrix Solution Spraying

Matrix solution spraying is a sample-preparation technique that applies a fine, uniform coating of a chemical matrix to a surface, often a tissue section, before mass spectrometric analysis. In matrix-assisted laser desorption/ionization, the matrix co-crystallizes with molecules in the sample and absorbs laser energy, promoting their desorption and ionization while helping preserve spatial information. In neuroscience, controlled spraying of matrix solution supports imaging mass spectrometry of brain tissue, enabling researchers to map the distributions of lipids, metabolites, peptides, and other biomolecules. Consistent coverage and crystal formation improve signal quality, molecular detection, and the interpretation of neurochemical patterns.

We present protocols to be used in the measurement of spray droplet size from agricultural nozzles used in both aerial and ground based agrochemical applications. These methods presented were developed to provide consistent and repeatable droplet size data both inter- and intra-laboratory, when using laser diffraction systems.

Topical Airway Anesthesia for Awake-endoscopic Intubation Using the Spray-as-you-go Technique with High Oxygen Flow

The topical anesthetic lidocaine was atomized using a high oxygen flow through the working channel of a flexible intubating endoscope to achieve topical airway anesthesia for awake endotracheal intubation. We prefer this modified spray-as-you-go technique for endoscopic intubation to classical bolus application because of higher patient satisfaction and better compliance.

Dithranol (DT; 1,8-dihydroxy-9,10-dihydroanthracen-9-one) has previously been reported as a MALDI matrix for tissue imaging of small molecules; protocols for the use of DT for the MALDI imaging of endogenous lipids on the surface of tissue sections by positive-ion MALDI-MS on an ultrahigh-resolution quadrupole-FTICR instrument are provided here.
