Mito-rfp Labeling

Mito-rfp labeling is a fluorescent protein-based method for visualizing mitochondria in living or fixed cells, making these energy-producing organelles easier to track in neuroscience research. The approach uses a red fluorescent protein fused to a mitochondrial targeting sequence, which directs the labeled protein to mitochondria and produces a detectable red signal under fluorescence microscopy. Researchers use this signal to examine mitochondrial distribution, morphology, transport, and dynamics in neurons and glial cells. Mito-rfp labeling can reveal changes associated with neuronal development, synaptic function, cellular stress, and neurodegenerative disease, supporting studies of how mitochondrial organization influences nervous-system health.

We describe a novel red fluorescent protein (RFP) reporter that is expressed specifically in the Drosophila eye. We detail a methodology for dissection of the eye imaginal disc and how this reporter can be used to aid in the dissection and identification of specific cell types in the developing eye.

A spectrofluorometric protocol for the measurement of the mitochondrial permeability transition pore opening in isolated mouse heart mitochondria is presented here. The assay involves the simultaneous measurement of mitochondria Ca2+ handling, mitochondrial membrane potential and mitochondrial volume. The procedure for obtaining high-quality and functional heart mitochondria is also described.

We describe a rapid and effective method for purification of mitochondria from the yeast Saccharomyces cerevisiae. This method enables the high-yield isolation of pure mitochondria that are essentially free of contamination by other organelles and retain their structural and functional integrity after their purification.
