Neuromuscular Function

Neuromuscular function describes how the nervous system communicates with skeletal muscles to generate movement, maintain posture, and regulate force. When a motor neuron fires, it releases acetylcholine at the neuromuscular junction, triggering an electrical signal in the muscle fiber that leads to calcium release, actin–myosin interaction, and contraction. Coordinated recruitment of motor units allows muscles to adjust strength and precision across different tasks. Studying neuromuscular function is central to neuroscience because it clarifies motor control and helps identify disruptions associated with conditions such as myasthenia gravis, motor neuron disease, peripheral neuropathy, and muscular disorders.

A functional assessment of the neuromuscular junction (NMJ) can provide essential information on the communication between muscle and nerve. Here we describe a protocol to comprehensively evaluate both the NMJ and muscle functionality using two different muscle-nerve preparations, i.e. soleus-sciatic and diaphragm-phrenic.

Assessment of Neuromuscular Function Using Percutaneous Electrical Nerve Stimulation

We present a protocol to assess changes in neuromuscular function. Percutaneous electrical nerve stimulation is a non-invasive method that evokes muscular responses. Electrophysiological and mechanical properties of these responses permit the evaluation of neuromuscular function from brain to muscle (supra-spinal, spinal and peripheral levels).

Assessing a Functional Neuromuscular Junction Via Simultaneous Optical Stimulation and Video Recording

2025

This video demonstrates a method to assess functional neuromuscular junctions. Using a bioreactor containing muscle tissue and motor neurons, along with fluorescent microscopy, the light-sensitive channels in the neurons are stimulated to generate electrical signals, causing muscle fiber contractions.

Drosophila Neuromuscular Junction (NMJ) Quantification: A Method to Assess Synaptic Morphology and Function

2023

The Drosophila neuromuscular junction (NMJ) is used as a model to study the morphology and function of synapses. This video describes important features that researchers quantify to characterize the NMJ. The example protocol demonstrates a software able to perform the quantification automatically.

Assessing Muscle Contraction with Neuromuscular Blocker Treatment

This video demonstrates assessing the muscle using cultured motor neurons, Schwann cells, and myotubes, followed by treatment with a neuromuscular blocker to inhibit contraction. Color-coded time-motion graphics show a color-coded time-motion graphic. Red indicates the fastest movement, and blue indicates the slowest.
