Plasmid Encoded Microrna

Plasmid-encoded microRNA is a molecular biology strategy in which circular DNA vectors carry sequences that direct cells to produce specific microRNAs, enabling targeted post-transcriptional regulation of gene expression. After plasmid delivery, a promoter drives transcription of a primary microRNA transcript, which is processed by Drosha and Dicer into a mature microRNA that joins the RNA-induced silencing complex and guides repression or degradation of complementary messenger RNA. In neuroscience, this approach allows researchers to manipulate genes in neuronal cultures and experimental models to investigate synaptic plasticity, neurodevelopment, and disease-related pathways. It also supports functional studies of candidate therapeutic targets and cellular mechanisms.

Plasmid Purification

2023

Plasmid purification is a technique used to isolate and purify plasmid DNA from genomic DNA, proteins, ribosomes, and the bacterial cell wall. A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules. When introduced into a host organism via transformation, a plasmid will be replicated, creating numerous copies of the DNA fragment under study. In this video, a step-by-step generalized procedure is described for how to perform plasmid purification.

Source: Laboratory of Jonathan Flombaum—Johns Hopkins University Long-term memory is a critical feature of human cognition, and it has been a prominent focus of research in experimental psychology. Many paradigms designed to tap long-term memory rely on asking participants to learn or study content, then test memory about that content. This is a good approach if one wants to understand how memory supports educational achievement, for example, where explicit study is part of the process. But,...
