Plasmid Vector Injection

Plasmid vector injection is a gene-delivery technique that introduces engineered circular DNA into selected cells or tissues, enabling expression of a gene of interest. In neuroscience, the injected plasmid typically contains a promoter, coding sequence, and regulatory elements; after cellular uptake and trafficking to the nucleus, transcription produces the encoded protein in targeted neural cells. This approach supports fluorescent reporter expression, manipulation of neuronal signaling or gene function, and analysis of circuit organization, providing a flexible platform for studying cellular mechanisms underlying brain function and disease.

(2018).In this video, a spinal and bulbar muscular atrophy or SBMA-affected mouse undergoes tail vein injection of recombinant AAV (Adeno-associated vectors) vectors carrying a plasmid encoding therapeutic microRNA. The vector produces microRNA that binds to mutant AR (Androgen receptor)-mRNA (messenger ribonucleic acid),...

Plasmid Purification

2023

Plasmid purification is a technique used to isolate and purify plasmid DNA from genomic DNA, proteins, ribosomes, and the bacterial cell wall. A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules. When introduced into a host organism via transformation, a plasmid will be replicated, creating numerous copies of the DNA fragment under study. In this video, a step-by-step generalized procedure is described for how to perform plasmid purification.
