Thrombolysis Protocol

A thrombolysis protocol is a structured clinical approach for using clot-dissolving medication to restore blood flow through an obstructed blood vessel, particularly in acute ischemic stroke. The treatment commonly relies on tissue plasminogen activator, which converts plasminogen into plasmin; plasmin then breaks down fibrin within the clot, allowing circulation to resume in threatened brain tissue. In neuroscience, protocols guide rapid patient assessment, brain imaging, eligibility screening, dosing, monitoring, and management of complications such as bleeding. Proper implementation can limit neuronal injury and improve functional recovery, while careful selection is essential because treatment benefits must be balanced against hemorrhagic risk.

To simulate thromboembolic stroke, emboli prepared from heterologous rat blood were injected into the middle cerebral artery, followed by administration of systemic thrombolysis for recanalization. This version of the model is optimized for use in a multi-laboratory network and supports testing multiple candidate therapeutics.

In-vitro thrombolysis assays have often struggled to replicate in-vivo conditions whether in the model thrombus being digested or in the environment in which thrombolysis is occurring. Herein, we explore how coupling the Chandler loop and Real-Time Fluorometric Flowing Fibrinolysis Assay (RT-FluFF) is used for high-fidelity, ex-vivo, clot lysis monitoring.

Neutrophils are among the first cells to arrive on the site of inflammatory immune response, and their functions and mechanisms have been studied extensively in vitro. We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.
