Sulfated Sucrose Polymer

Sulfated sucrose polymer is a negatively charged, chemically modified carbohydrate used in pharmacology to protect damaged gastrointestinal mucosa. In the acidic stomach, its sulfate groups promote binding to positively charged proteins at ulcerated or inflamed tissue, forming an adherent protective layer that limits exposure to acid, pepsin, and bile salts. This localized barrier supports healing in peptic ulcers and other gastric or duodenal lesions while producing minimal systemic effects because the polymer is poorly absorbed. Studying its structure and acid-dependent activity illustrates how polymer chemistry can guide drug action at the site of injury.

In this video, we demonstrate a procedure for combined Alcian blue and silver staining of sulfated glycosaminoglycan oligosaccharide fragments. This combined staining technique significantly amplifies the bands of glycosaminoglycan polymers of different lengths in polyacrylamide gels.

Here, we present a protocol for the synthesis of placental chondroitin sulfate A binding peptide (plCSA-BP)-conjugated lipid-polymer nanoparticles via single-step sonication and bioconjugate techniques. These particles constitute a novel tool for the targeted delivery of therapeutics to most human tumors and placental trophoblasts to treat cancers and placental disorders.
