Sulfhydryl-containing compounds are molecules that carry a thiol group (-SH), a chemically reactive feature that influences redox balance, protein structure, and drug activity. The sulfur atom can undergo oxidation to form disulfide bonds, participate in nucleophilic reactions, or bind reactive electrophiles, allowing these compounds to modify enzymes, receptors, and other biological targets. In pharmacology, thiol chemistry helps explain the actions of drugs such as captopril and acetylcysteine, including target binding, antioxidant effects, and detoxification of reactive metabolites. Understanding these mechanisms supports drug design, adverse-effect assessment, and therapies that restore or protect cellular sulfhydryl groups.

A method to analyze the solvent accessibility of the thiol group of cysteine residues of Maize rayado fino virus (MRFV)-virus-like particles (VLPs) followed by a peptide cross-linking reaction is described. The method takes advantage of the availability of several chemical groups on the surface of the VLPs that can be targets for specific reactions.

This video demonstrates co-immunoprecipitated protein complexes on a PVDF membrane for protein-protein interaction analysis. The reduced proteins from complexes were treated with trypsin to cleave the individual proteins at the smaller peptides, then, the remaining peptides were extracted from the PVDF membrane. The pulled peptide derived from both proteins in the complex was then dried and resuspended in a low concentration of formic acid for further analysis.
