Evolutionary Relationships

Los seres humanos han clasificado y organizado organismos biológicos durante miles de años. Originalmente, ordenaba principalmente los objetos necesarios para la supervivencia. A medida que avanzaba la historia de la humanidad, también lo hacía la habilidad y el detalle de estas clasificaciones. Hoy en día, los científicos comúnmente construyen árboles llamados dendrogramas para dar representaciones visuales de estas divisiones y grupos. Esta forma particular de dendrograma, el cladograma, visualiza las relaciones cladísticas entre las especies, de modo que las puntas del árbol representan las especies y las ramas muestran cómo se relacionan entre sí. Por ejemplo, aquí el chimpancé y el oso están más estrechamente relacionados entre sí y comparten características más comunes que cualquiera de ellos con el pez luna. Los lugares donde se unen las ramas se denominan nodos y denotan ancestros comunes para las especies que siguen. Un segundo tipo importante de dendrograma es el filograma. Por lo tanto, cuanto más larga es la rama, más tiempo ha pasado desde que la especie divergió de su último ancestro común.
Los dendrogramas se construyeron simplemente analizando la morfología de los organismos. Con el advenimiento de la tecnología moderna, la comparación del ADN también se ha convertido en una forma común de construir árboles. El ADN está formado por nucleótidos asociados a una de cuatro bases diferentes. Adenina, guanina, citosina o timina. El orden de estas bases es el código de ADN. Este código se transmite de padres a hijos. En consecuencia, si nos fijamos en una sola especie como los humanos, hay un alto grado de similitud en nuestro código genético, alrededor del 99,9%. También compartimos parte de nuestro código de ADN con otras especies, como los chimpancés y los ratones, pero el grado de similitud general entre nuestro ADN y el suyo es muy diferente. Esto significa que podemos crear árboles, que agrupan especies en función de las similitudes o diferencias entre sus códigos genéticos. Este campo de análisis, que combina estadística, modelado matemático y ciencias de la computación, se conoce como bioinformática. Para comparar las secuencias de ADN, los investigadores a menudo utilizan una herramienta bioinformática llamada Herramienta Básica de Búsqueda de Alineación Local, o BLAST, que fue creada y es mantenida por el Centro Nacional de Información Biotecnológica.
En este laboratorio, primero creará un cladograma de animales utilizando información morfológica y, a continuación, colocará una especie fósil en este cladograma en función de su morfología. <h4>Tipos de dendrograma</h4>
Un tipo de dendrograma, llamado cladograma, representa las relaciones genealógicas hipotéticas entre especies con las puntas (u hojas) del árbol que representan una especie y las ramas que muestran cómo las especies se relacionan entre sí. Un tipo de árbol un poco más complicado, llamado filograma, difiere de un cladograma en que las ramas que conducen a la especie son de diferentes longitudes. La longitud de una rama en este tipo de árbol representa el grado de cambio entre especies: cuanto más larga es la rama, más tiempo hace que las especies divergen de un ancestro común. En ambos tipos de árboles, el ancestro común de un grupo de especies se indica mediante un nudo, que es el punto donde se unen una serie de ramas. Las especies que están más estrechamente relacionadas entre sí (más recientemente compartidas con un ancestro común) se ubicarán más cerca del nodo. Las dos especies que comparten un nodo se denominan grupo hermano1.
<h4>Comprensión de las relaciones evolutivas mediante el uso de datos genéticos</h4>
Históricamente, los cladogramas se construían comparando la morfología (estructura física) de los organismos. Este método todavía se practica, pero las técnicas se han modernizado para incluir la comparación de secuencias de ADN (ácido desoxirribonucleico) entre especies. El uso del ADN para la construcción de árboles tiene varias ventajas en comparación con confiar únicamente en la morfología, incluida la capacidad de calcular una estimación de cuánto tiempo hace que las diferentes especies compartieron un ancestrocomún. El ADN se transmite de padres a hijos en unidades hereditarias llamadas genes. La bioinformática son las herramientas utilizadas por los biólogos para analizar grandes conjuntos de datos utilizando una combinación de informática, modelado matemático y estadística. Una de estas herramientas se llama BLAST (Basic Local Alignment Search Tool), que se puede utilizar para buscar rápidamente el genoma completo de cualquier especie que esté disponible en la base de datos del NCBI (Centro Nacional de Información Biotecnológica)2. La base de datos del NCBI combina varias bases de datos diferentes que contienen diferentes tipos de información de secuencias de ADN. El proceso de una búsqueda BLAST incluye algoritmos informáticos complejos, pero básicamente, BLAST alinea las secuencias de cada base de nucleótidos de una secuencia de ADN enviada (conocida como secuencia de consulta) con las secuencias de la base de datos que más se acercan a ella. Las secuencias de ADN que se encuentren se enumerarán en orden de similitud con la secuencia en cuestión y, por lo tanto, serán de especies estrechamente relacionadas con las especies que contienen el gen de consulta. Esta comparación puede o no representar la relación evolutiva real entre las especies porque los genes evolucionan a diferentes ritmos. Además, los genomas a veces contienen más de una instancia de una secuencia similar.
La comparación de las secuencias de ADN de los genes es valiosa más allá de la consideración de las relaciones evolutivas. Con frecuencia, los genes se identifican en organismos modelo, como la mosca de la fruta, Drosophila melanogaster, o el ratón3. Como parte integral del estudio de un gen, la función de su producto se identifica y analiza comúnmente. Si un investigador está interesado en estudiar esa función en un organismo diferente (humanos, por ejemplo), BLAST u otras herramientas bioinformáticas se pueden utilizar para encontrar genes candidatos en función de sus similitudes con los genes de función conocida de organismos modelo.
Los genes humanos también se pueden utilizar como punto de partida para encontrar homólogos en organismos modelo. De hecho, la investigación de enfermedades humanas depende en gran medida de esto. Una vez que se identifica un gen humano de interés, los ratones pueden ser manipulados genéticamente para que el gen homólogo sea alterado o "eliminado", creando un modelo de la enfermedad humana que se puede estudiar para comprender y tratar la enfermedad. Hay muchas de estas cepas de ratones disponibles actualmente. Por ejemplo, hay un modelo de ratón para la fibrosis quística (FQ) humano llamado ratón knockout Cftr y otro modelo de aterosclerosis, llamado Apoe knockout3.

DNA evidence helps scientists map evolutionary relationships between living things. Early classification began with Aristotle in the 4th century BC and was later improved during the Renaissance and by Carolus Linnaeus in the mid-1700s. These systems grouped species by physical similarity, such as the presence or absence of a backbone in vertebrates and invertebrates. Traits like a backbone are called synapomorphies, which are shared traits that likely came from a common ancestor.

Physical traits are still useful, but they also have limits. To show how species are related, scientists build dendrograms, or branching diagrams. A cladogram is one type of dendrogram. It shows hypothetical family-like relationships, with the tips representing species and the branches showing how they are connected.

A phylogram is a more detailed tree. Its branch lengths are not the same. The length of a branch shows the amount of change between species, so a longer branch means more time since two species diverged from a common ancestor. In both cladograms and phylograms, a node marks the point where branches meet. Species that share the most recent common ancestor are closest to that node and are called sister groups.

Scientists used to make cladograms by comparing morphology, or body structure. That method is still used, but it now includes DNA sequence comparison. DNA evidence can also help estimate how long ago species shared a common ancestor. This is especially useful for understanding evolutionary history, although it does not always work for extinct organisms because DNA is usually found in soft tissue, and soft tissue is rarely preserved in fossils.

Genes carry DNA from parents to offspring. The nucleotide sequence in genes uses the bases A, G, C, and T. These sequences are often similar in different species because of common ancestry. Researchers can align them to build trees. Species with more similar sequences are placed closer together, while species with less similarity are placed farther apart.

Bioinformatics tools help scientists work with large DNA datasets. These tools combine computer science, math, and statistics. One example is BLAST, or Basic Local Alignment Search Tool. BLAST searches the NCBI database, which brings together several databases of DNA sequence information. It compares a query sequence against sequences in the database and lists the closest matches first.

A BLAST result can suggest which species are closely related to the source of the query gene. However, this comparison may not always show the exact evolutionary relationship. Genes can evolve at different rates, and genomes can contain more than one similar sequence. Even so, DNA comparisons are valuable for more than tree building. Scientists often use genes first found in model organisms such as the fruit fly Drosophila melanogaster or the mouse to study gene function in other species, including humans. Human genes can also be used to find homologs in model organisms. This approach is important in human disease research. For example, researchers can knock out a mouse version of a human gene to create disease models such as the Cftr knockout mouse for cystic fibrosis or the Apoe knockout mouse for atherosclerosis.
