Enzyme Activity

¿Qué es un catalizador? Es una sustancia que permite que una reacción ocurra más rápido, utilizando menos energía. Si trazaras un gráfico de la energía necesaria para una reacción biológica no catalizada, se vería algo así. Esta altura representa la energía de activación, que es la cantidad mínima de energía necesaria para que la reacción continúe. Las enzimas, que funcionan como catalizadores, proporcionan una vía de reacción alternativa, con una energía de activación más baja. 

Entonces, ¿cómo funcionan realmente las enzimas? La mayoría de las enzimas son proteínas. La parte de su estructura que interactúa con las moléculas que catalizan se llama sitio activo, que tiene afinidad solo por ciertos sustratos y no se une a otros. Cuando los sustratos correctos ingresan al sitio activo, la enzima cambia su forma para lograr una configuración de mayor afinidad. Este estado de transición facilita la reacción y convierte los reactivos en productos. Resolvieron este problema encurtiendo frutas y verduras ricas en vitamina C. Por lo tanto, la destrucción de las enzimas microbianas y el almacenamiento de una coenzima esencial jugaron un papel importante en el éxito de las expediciones largas. 

En este laboratorio, puede cuantificar cómo las diferentes condiciones afectan la función de la enzima, evaluando la velocidad de reacción, la cantidad de producto producido, por unidad de tiempo. Primero medirá la velocidad de reacción de referencia a una temperatura ambiente estándar y un pH neutro. La tasa aumentará al principio hasta que todas las moléculas de la enzima estén completamente saturadas con el sustrato. Y luego, la catálisis alcanzará un nivel constante a medida que las enzimas estén ocupadas continuando el ciclo de unión al sustrato, catálisis y liberación. Una vez que se ha establecido la velocidad de reacción de referencia, se puede utilizar como punto de referencia para otras condiciones. La enzima con la que trabajará en este laboratorio es la peroxidasa de nabo. Evaluará su actividad bajo valores variables de temperatura y pH.

<h4>Catalizadores biológicos</h4>

Todos los organismos vivos realizan continuamente numerosas reacciones bioquímicas para mantener su presencia. La mayoría de estas reacciones requieren una entrada de energía para comenzar, que se denomina energía de activación. Los catalizadores son sustancias químicas que disminuyen la energía de activación. Aunque los catalizadores facilitan una reacción química, no son consumidos por ella. Esto significa que un catalizador puede facilitar una reacción química específica una y otra vez en sucesión, aumentando la velocidad de esa reacción.

Las enzimas son catalizadores biológicos que aumentan la velocidad y la eficiencia de las reacciones bioquímicas. La mayoría de las enzimas son proteínas, pero ciertas moléculas de ácido ribonucleico también tienen propiedades catalíticas. Cada tipo de enzima solo puede unirse a uno o unos pocos sustratos específicos, por lo tanto, existe una gran diversidad de enzimas, todas con funciones distintas. La reacción específica de una enzima depende de sus sitios activos, que tienen formas que coinciden estrechamente con la forma de sus sustratos. Al unirse al sustrato, la enzima cambia ligeramente de forma de una manera que permite que la reacción química ocurra más fácilmente, reduciendo así la energía de activación de la reacción. Una vez que se produce la reacción, la enzima libera los productos y vuelve a su forma original, lo que le permite repetir el proceso de nuevo.

<h4>Enzimas catabolíticas versus anabólicas</h4>

La mayoría de las reacciones enzimáticas se pueden clasificar como catabólicas y anabólicas, que tienen funciones opuestas. Las enzimas que median las reacciones catabólicas descomponen las sustancias más grandes en múltiples productos. Una enzima catabólica bien conocida es la lactasa, que divide el disacárido lactosa de los productos lácteos en monosacáridos galactosa y glucosa. Los seres humanos tienen grandes cantidades de lactasa en su intestino delgado después del nacimiento, sin embargo, muchas personas pierden más del 90% de su lactasaen la primera infancia. Esta reducción disminuye la capacidad de digerir la lactosa de una manera dependiente de la edad, lo que se conoce como intolerancia a la lactosa2.

Las enzimas anabólicas combinan múltiples sustancias en un solo producto. Por ejemplo, la enzima ADN polimerasa sintetiza moléculas de ADN uniendo nucleótidos. Del mismo modo, la enzima glucógeno sintasa conecta las moléculas de glucosa para generar glucógeno. La insulina regula la actividad de la glucógeno sintasa, por lo tanto, los pacientes diabéticos con niveles bajos o nulos de insulina no pueden sintetizar glucógeno sin tratamiento con insulina3.

<h4>Factores ambientales</h4>

La forma y la carga del sitio activo de una enzima se adaptan a la reacción química específica que cataliza, por lo tanto, la actividad de la enzima está influenciada por factores que afectan su sitio activo. Por lo tanto, las enzimas requieren condiciones específicas para funcionar a niveles óptimos y, dependiendo de su función y ubicación, estas condiciones pueden variar mucho. Estos factores incluyen el pH y la temperatura. Por ejemplo, los sitios activos de las enzimas a menudo contienen aminoácidos ácidos o básicos. Las desviaciones del pH óptimo alteran las cargas en el sitio activo, lo que dificulta la interacción de los sustratos con la enzima. Esta propiedad se ha utilizado durante siglos para conservar los alimentos mediante el encurtido, que reduce el pH del alimento en escabeche a un nivel que inhibe eficazmente la actividad enzimática microbiana. Del mismo modo, cada enzima funciona en un rango de temperatura óptimo. Si la temperatura cambia por debajo o por encima de este rango, altera la forma del sitio activo, impidiendo que interactúe efectivamente con sus sustratos. En la mayoría de los casos, volver a la temperatura óptima restaura la forma y función originales de la enzima, sin embargo, las temperaturas demasiado altas pueden desnaturalizar o dañar irreversiblemente la estructura de la enzima. Los extremófilos, u organismos que se encuentran en condiciones extremas que son letales para la mayoría de las formas de vida, tienen enzimas capaces de funcionar en esos extremos. Por ejemplo, Thermus aquaticus es una bacteria que se encuentra en las aguas termales y en los respiraderos hidrotermales que alcanzan temperaturas a las que la mayoría de las proteínas se desnaturalizarían. La polimerasa Taq es una ADN polimerasa termoestable que se encuentra en T. aquaticus, lo que ha permitido el desarrollo de la reacción en cadena de la polimerasa (PCR), un método de laboratorio ampliamente utilizado para la replicación rápida del ADN. La PCR requiere altas temperaturas para separar las hebras de la doble hélice de ADN para ser replicadas. La polimerasa Taq permanece estable a estas altas temperaturas, mientras que la mayoría de las enzimas se degradan. La PCR alterna entre altas temperaturas que rompen el ADN en monocatenarios y temperaturas más bajas que permiten a la polimerasa construir nuevo ADN bicatenario.

<h4>Factores moleculares que influyen en la actividad enzimática</h4>

Además de los rangos específicos de pH y temperatura, algunas enzimas funcionan mejor en presencia de otras moléculas o iones. Las coenzimas son biomoléculas que se unen al sitio activo de la enzima y participan en la catálisis sin ser consumidas por la reacción. Las coenzimas a menudo transportan electrones, átomos específicos y grupos funcionales, y por lo tanto actúan como portadores intermediarios. Las vitaminas del grupo B actúan como coenzimas en numerosas reacciones bioquímicas. Por ejemplo, la coenzima A (coA), que es la forma activa del ácido pantoténico, es requerida por el 4% de todas las enzimasde los mamíferos. Los cofactores a menudo se clasifican como sustancias inorgánicas, como iones, que son necesarias para la función enzimática. Por ejemplo, todas las enzimas ADN polimerasa requieren Mg2+ o Mn2+ para la replicación del ADN, así como para la escisión de nucleótidos incorporados incorrectamente5.

También hay otras moléculas que no son necesarias para que una enzima funcione, pero que son capaces de alterar la actividad enzimática. Las sustancias que aumentan la actividad enzimática se denominan activadores, mientras que otras que disminuyen la actividad enzimática se denominan inhibidores. Estas moléculas se unen a las enzimas en el sitio activo o en otros lugares de la enzima conocidos como sitios alostéricos. A veces, estas moléculas pueden unirse a la ubicación exacta en el sitio activo como sustrato y, por lo tanto, competir con el sustrato por la unión de la enzima. Otras, especialmente las sustancias que se unen a los sitios alostéricos, pueden alterar la función de la enzima sin bloquear la unión del sustrato al sitio activo y, por lo tanto, no son competitivas. Además, la unión de los inhibidores puede ser reversible o irreversible. Cuando la interacción es reversible, la enzima puede reanudar su función una vez que el inhibidor se desprende. Sin embargo, cuando la unión es irreversible, el inhibidor nunca se separa de la enzima y la enzima se daña irreparablemente y necesita ser reemplazada. Sin embargo, los inhibidores enzimáticos también son útiles. Por ejemplo, los inhibidores se han utilizado como antibióticos para inhibir el crecimiento bacteriano y como quimioterapéuticos para inhibir la división descontrolada de las célulascancerosas 6.

<h4>Medición de la función enzimática</h4>

Un método utilizado por los científicos para estudiar los efectos de diferentes condiciones en la función de la enzima es cuantificar la velocidad de reacción, que es la rapidez con la que la enzima cataliza la reacción. Por lo tanto, una velocidad de reacción más alta indica que la enzima está catalizando reacciones de manera más eficiente, mientras que una tasa más baja significa que está funcionando de manera menos eficiente. Para determinar la velocidad de reacción, la velocidad de reacción se calcula midiendo la cantidad de producto producido por unidad de tiempo. Esto se hace midiendo la concentración de producto comenzando justo al comienzo de la reacción, cuando la concentración de producto aumenta linealmente, hasta que la enzima está completamente saturada con el sustrato y la catálisis alcanza un nivel constante. Una vez que se determina la velocidad de reacción de referencia, se puede utilizar como punto de referencia, o control, al comparar entre una variedad de tratamientos. Podemos evaluar las velocidades de reacción como medidas relativas o absolutas. Una velocidad de reacción relativa es una medida comparativa que muestra las diferencias entre dos o más condiciones, como los tratamientos en un solo experimento, pero no da un valor para lo que se está midiendo. Una velocidad de reacción absoluta se basa en una medida cuantitativa del progreso de la reacción. Una manera fácil de entender la diferencia entre las tasas de reacción relativas y absolutas es considerar la medición de la velocidad de un coche en una carrera. Un método relativo compararía evaluando la clasificación de los coches en la línea de meta, mientras que la tasa absoluta evaluaría y compararía la velocidad de los coches individuales en millas por hora.

Dado que las enzimas se usan tan ampliamente, los investigadores han comenzado a buscar formas de crear y modificar enzimas para satisfacer necesidades específicas, lo que se conoce como ingeniería de proteínas. La tecnología de ADN recombinante permite cambiar las secuencias de aminoácidos para alterar la forma de una enzima o la afinidad del sitio activo. Esto se ha utilizado para crear pruebas diagnósticas altamente sensibles para diversas afecciones, así como para los esfuerzos de biorremediación y las herramientas de gestión de residuos7.

factors that shape enzyme function

Enzymes are biological catalysts that help living things run biochemical reactions faster. They lower activation energy, which is the energy needed to start a reaction. Enzymes are not used up in the process, so they can keep working again and again. Most enzymes are proteins, although some ribonucleic acid molecules also have catalytic activity.

Each enzyme usually binds only one or a few specific substrates. This selectivity depends on the enzyme’s active site, which fits its substrate like a matching shape. When the substrate binds, the enzyme changes shape slightly and makes the reaction easier. After the products form, the enzyme releases them and returns to its original shape.

Enzyme reactions are often grouped as catabolic or anabolic. Catabolic enzymes break larger molecules into smaller products. Lactase is a catabolic enzyme that splits lactose in dairy products into galactose and glucose. Many people lose most of their lactase by early childhood, which can lead to lactose intolerance. Anabolic enzymes build larger molecules from smaller parts. DNA polymerase joins nucleotides to make DNA, and glycogen synthase links glucose molecules to form glycogen. Insulin helps regulate glycogen synthase, so people with little or no insulin may need treatment to make glycogen normally.

Enzyme activity depends on conditions such as pH and temperature. Many active sites contain acidic or basic amino acids, so changes in pH can alter charge and make substrate binding harder. Pickling uses this idea by lowering pH and slowing microbial enzyme activity. Temperature also matters because each enzyme has an optimal range. Low or moderate shifts may slow the enzyme, but high temperatures can denature it and cause lasting damage. Cooking meat and refrigerating food both reduce harmful microbial activity in this way. Extremophiles live in harsh environments and have enzymes that still work there. Thermus aquaticus is one example, and its Taq polymerase stays stable in the high heat used for PCR, a method that copies DNA by cycling between hot and cooler temperatures.

Some enzymes also need coenzymes or cofactors. Coenzymes are biomolecules that help the reaction and are not consumed. They often carry electrons, atoms, or functional groups, and many B vitamins serve this role. Coenzyme A, the active form of pantothenic acid, is needed by many mammalian enzymes. Cofactors are usually inorganic substances, such as ions. DNA polymerase, for example, requires Mg2+ or Mn2+ for DNA replication and for correcting some mistakes.

Other molecules can change enzyme activity without being required for the reaction. Activators increase activity, while inhibitors decrease it. These molecules may bind at the active site or at allosteric sites, which are different locations on the enzyme. Some inhibitors compete with the substrate for the active site, while others act in a non-competitive way by binding elsewhere. Inhibitor binding can also be reversible or irreversible. Reversible inhibitors can detach and let the enzyme work again, but irreversible inhibitors permanently damage the enzyme. Scientists use enzyme inhibitors as antibiotics and as chemotherapeutic agents to limit cancer cell division.

Scientists measure enzyme function by looking at reaction rate, or how fast product is made. A faster rate means the enzyme is working more efficiently. Reaction velocity is measured by tracking product concentration over time, usually from the start of the reaction until the enzyme becomes saturated with substrate and the rate levels off. Researchers use that baseline as a control when comparing different treatments. Reaction rates can be reported as relative or absolute values. Relative rates compare conditions, while absolute rates give a direct quantitative measure, much like comparing race rankings versus miles per hour.

Because enzymes are so useful, researchers also engineer them for special tasks. Protein engineering uses recombinant DNA technology to change amino acid sequences and adjust enzyme shape or active site binding. This approach has helped create sensitive diagnostic tests, as well as tools for bioremediation and waste management.
