El cloruro de sulfonilo reacciona con las aminas primarias y secundarias para producir las sulfonamidas sustituidas correspondientes, pero no reacciona con las aminas terciarias ya que la amina no tiene protones N-H.
El grupo sulfonilo que retira electrones hace que el protón N-H de sulfonamida sea lo suficientemente ácido como para reaccionar con una base y producir una sal soluble en agua.
Para distinguir entre los tipos de aminas, las respectivas mezclas de aminas en base acuosa se tratan con cloruro de bencenosulfonilo, también conocido como reactivo de Hinsberg. A esto se le llama la prueba de Hinsberg.
Una amina primaria reacciona con el cloruro de bencenosulfonilo para formar un precipitado de sulfonamida N-sustituido que se disuelve en la base para generar una solución clara de una sal soluble en agua. Además, la adición de un ácido precipita la sulfonamida N-sustituida.
De manera similar, las aminas secundarias reaccionan con el cloruro de sulfonilo para dar sulfamidas N,N-disustituidas insolubles en agua. Dado que no tiene protón ácido, permanece insoluble en la base añadida.
Las aminas terciarias no reaccionan con el reactivo de Hinsberg y, por lo tanto, no se forman precipitados.

The Hinsberg test identifies primary, secondary, and tertiary amines by their reaction with benzenesulfonyl chloride, also called the Hinsberg reagent. It is named after Oscar Hinsberg. The test uses excess aqueous base, and the mixture is then acidified to show different results for each amine type.

A primary amine reacts with the Hinsberg reagent to form an N-substituted benzenesulfonamide. The sulfonyl group pulls electron density away and makes the N-H proton acidic. Because of this, base can remove that proton and form a water-soluble salt. When the solution is acidified again, the N-substituted benzenesulfonamide precipitates.

A secondary amine gives an insoluble N,N-disubstituted sulfonamide after reaction with benzenesulfonyl chloride. This product has no N-H proton, so it does not react with the base. A tertiary amine does not react with the Hinsberg reagent at first. After acidification, however, the tertiary amine is protonated and becomes water-soluble.
