Hypotonic Swelling

Hypotonic swelling is the increase in cell volume that occurs when cells are placed in a solution with a lower solute concentration than their cytoplasm, making it an important example of osmotic regulation in biochemistry. Water moves across the plasma membrane by osmosis toward the higher internal solute concentration, and continued influx can stretch the membrane, disrupt cellular function, or cause lysis when membrane limits are exceeded. Researchers use hypotonic swelling to assess membrane permeability and integrity, study cellular volume control, and induce controlled cell disruption for biochemical analyses, including the release of intracellular components.

Pf determination in spherical plant cell protoplasts presented here involves protoplasts isolation and numerical analysis of their initial rate of volume change as a result of an osmotic challenge during constant bath perfusion.
