Nucleoside Nucleotide Quantification

Nucleoside nucleotide quantification is the measurement of nucleosides and nucleotides in biological or biochemical samples, providing insight into cellular energy status, nucleic acid metabolism, and signaling. Analytical workflows typically extract these polar metabolites, separate them by high-performance liquid chromatography, and identify or measure them using ultraviolet detection or mass spectrometry against calibration standards. Careful control of sample handling, extraction conditions, and internal standards helps preserve unstable compounds and improve accuracy. In biochemistry, these measurements support studies of ATP and other energy carriers, metabolic pathway activity, enzyme function, cellular stress, and responses to drugs or genetic changes.

2025

This video demonstrates the utility of digital PCR in a chip-in-a-tube format for the detection of single nucleotide variations in DNA samples. A single PCR reaction is partitioned into chambers that act as independent PCR reactions, and the detection of fluorescence signals from the amplified targets in the chambers is used to compute the frequency of the variant allele in the sample.

(2017)This video demonstrates the infection of stationary-phase mammalian cells with HSV-1 (Herpes Simplex Virus-1), followed by replication of the viral genome using host and viral replication machinery. A synthetic nucleoside analog is incorporated into newly synthesized viral DNA to label replicating genomes, which are subsequently isolated for...
