Streptavidin Bead Capture

Streptavidin bead capture is a biochemical technique that isolates biotin-labeled molecules from complex samples through the exceptionally strong interaction between biotin and streptavidin. In this method, streptavidin-coated magnetic or agarose beads bind biotinylated proteins, nucleic acids, or other targets, allowing unbound components to be removed by washing before the captured material is recovered or analyzed. The approach supports affinity purification, pull-down assays, target enrichment, and biomolecular interaction studies. Because the interaction is highly selective and stable, streptavidin bead capture improves sample specificity and enables sensitive analysis of molecular complexes in research and diagnostic workflows.

In this article, a simple, quantitative, liquid phase affinity capture assay is presented. It is a reliable technique based on the interaction between magnetic beads and tagged proteins (e.g. nanobodies) on one hand and the affinity between the tagged protein and a second, labeled protein (e.g. poliovirus) on the other.

In this video, we demonstrate an immunoprecipitation assay, called the VLP capture assay, to isolate antigen-displaying virus-like particles, or VLPs, from a sample. The antigen binds with specific antibodies bound to magnetic beads, forming an immobilized antigen-antibody complex that is later separated and stored for further analysis.

Described is a two-step labeling process using β-glucosyltransferase (β-GT) to transfer an azide-glucose to 5-hmC, followed by click chemistry to transfer a biotin linker for easy and density-independent enrichment.
