Trna Ligase Co-expression

tRNA ligase co-expression is a biochemical strategy in which a tRNA ligase is produced alongside a target tRNA or other relevant protein to promote efficient tRNA maturation or repair. The ligase recognizes compatible tRNA termini, brings them together, and catalyzes phosphodiester-bond formation, restoring a continuous tRNA backbone after precursor processing or cleavage. In recombinant systems, coordinated expression can improve the availability and functionality of engineered tRNAs, support studies of tRNA splicing and RNA repair, and help analyze enzyme specificity. This approach is useful for investigating RNA metabolism and for developing systems that depend on reliable tRNA production and modification.

This protocol describes the preparation of a synthetic tRNA substrate for the Entamoeba histolytica DNA/tRNA methyltransferase 2 (Dnmt2) homolog Ehmeth and the measure of its methyltransferase activity. This experimental approach can be used for investigating the activity of other Dnmt2 proteins.
