Direct Culture Method

The Direct Culture Method is a laboratory technique that places a biological sample directly into a controlled culture system to maintain, grow, or assess its cells without an intermediate isolation or expansion step. In bioengineering, the sample is supplied with an appropriate medium and physical environment, allowing cells to attach, survive, proliferate, or organize while researchers monitor changes over time. This approach can preserve interactions among cells and surrounding material that may be lost during extensive processing, making it useful for evaluating engineered tissues, biomaterials, and cellular responses. Direct culture supports rapid screening and biologically relevant experiments, although sample variability and contamination require careful control.

We introduce three methods of direct culture, direct exposure culture, and exposure culture for evaluating the in vitro cytocompatibility of biodegradable implant materials. These in vitro methods mimic different in vivo cell-implant interactions and can be applied to study various biodegradable materials.

In this video, we demonstrate all-in-one CRISPR-Cas9 based genome editing in cultured cells where Cas9 and sgRNA are provided as a single plasmid construct to the cells. The CRISPR-Cas9 system and desired gene to be inserted was introduced in cells through electroporation technique to facilitate successful gene editing.
