Episomal Vectors

Episomal vectors are extrachromosomal DNA molecules engineered to carry and express genetic cargo without integrating into a host cell’s chromosomes, making them useful for controlled and potentially reversible gene delivery. They function by entering cells, directing transcription from their regulatory elements, and, in some systems, using viral origins and maintenance proteins to support episome replication and persistence; without these mechanisms, dilution during cell division can reduce expression. In bioengineering, episomal vectors support transient protein production, cellular reprogramming, genome engineering, and cell therapy research where limiting permanent genomic alteration is important.

Generation of Integration-free Induced Pluripotent Stem Cells from Human Peripheral Blood Mononuclear Cells Using Episomal Vectors

This protocol describes a detailed method for efficient generation of integration-free iPSCs from human adult peripheral blood cells. With the use of four oriP/EBNA-based episomal vectors to express the reprogramming factors, KLF4, MYC, BCL-XL, or OCT4 and SOX2, thousands of iPSC colonies can be obtained from 1 mL of peripheral blood.

We describe the production strategy of integrase-deficient lentiviral vectors (IDLVs) as vehicles for delivering CRISPR/Cas9 to cells. With an ability to mediate quick and robust gene editing in cells, IDLVs present a safer and equally effective vector platform for gene delivery compared to integrase-competent vectors.
