Notch Ligand Presentation

Notch ligand presentation is the controlled display of membrane-associated Delta-like or Jagged ligands to regulate Notch signaling between neighboring cells, an important strategy for directing cell fate in bioengineering. In trans, ligand binding to a Notch receptor on an adjacent cell, together with ligand endocytosis and mechanical force, exposes a cleavage site that enables proteolytic release of the Notch intracellular domain; this domain enters the nucleus and alters target-gene transcription. Engineers can present ligands on living feeder cells, engineered cell membranes, surfaces, or biomaterials while tuning density, mobility, spacing, and duration. These systems help model tissue organization and control stem-cell maintenance, differentiation, and regenerative design.

Complexity of in vivo systems makes it difficult to distinguish between the activation and inhibition of Notch receptor by trans- and cis-ligands, respectively. Here, we present a protocol based on in vitro cell-aggregation assays for qualitative and semi-quantitative evaluation of the binding of Drosophila Notch to trans-ligands vs cis-ligands.
