Potato Dextrose Agar

Potato Dextrose Agar (PDA) is a nutrient-rich solid culture medium used primarily to grow and maintain fungi, yeasts, and some other microorganisms in laboratory settings. It combines potato extract and dextrose as carbon and energy sources with agar, which forms a stable gel; its mildly acidic environment generally favors fungal growth over many bacteria. In bioengineering, PDA supports the isolation, cultivation, and visual assessment of fungal strains used in biotechnology, enzyme production, fermentation, plant-microbe studies, and biomaterial research. Consistent preparation and incubation conditions help researchers compare colony morphology, growth rates, and contamination across experiments.

This protocol describes a high through put colorimetric method that relies on the formation of a complex between iodine and chains of glucose molecules in starch. Iodine forms complexes with both amylose and long chains within amylopectin. After the addition of iodine to a starch sample, the maximum absorption of amylose and amylopectin occurs at 620 and 550 nm, respectively. We present a detailed protocol for potato virus X (PVX)-based microRNA silencing (VbMS) system to functionally characterize endogenous microRNAs (miRNAs) in potato. Target mimic (TM) molecules of miRNA of interest are integrated into the PVX vector and transiently expressed in potato to silence the target miRNA or miRNA family.

We describe a qualitative assay for yeast adhesion and agar invasion as a measure of invasive and pseudohyphal differentiation. This simple assay can be used to assess the invasive phenotype of various mutants as well as the effects environmental cues and signaling pathways on yeast differentiation.
