Extracellular Fluid Isolation

Extracellular fluid isolation is the process of separating fluid outside cells from cellular material in a biological sample, enabling direct study of its dissolved proteins, metabolites, ions, and signaling molecules. The sample is typically collected under conditions that limit contamination, then clarified by filtration or differential centrifugation to remove cells and debris while retaining soluble components. In biology, isolated extracellular fluid supports analysis of cell communication, tissue homeostasis, disease-related changes, and responses to treatment. By linking fluid composition with physiological state, the method helps researchers characterize microenvironments and identify measurable biomarkers for basic and translational research.

The model organism C. elegans uses pseudocoelomic fluid as a passive circulatory system. Direct assay of this fluid has not been previously possible. Here, we describe two extracellular vesicle isolation protocols, ultrafiltration centrifugation and ultracentrifugation with density gradient centrifugation, to isolate extracellular vesicles from murine bronchoalveolar lavage fluid samples. The extracellular vesicles derived from murine bronchoalveolar lavage fluid by both methods are quantified and characterized.

Characterizing Extracellular Vesicles from Biological Fluids

Here, we demonstrate the characterization method for extracellular vesicles (EVs) collected from biological fluids, such as tears and saliva, of human subjects. The scanner used in this method is capable of detecting the phenotype, size, and total particle count of EVs from 1 µL of the sample.
