Immune Tolerance Loss

Immune tolerance loss is the failure of the immune system to remain unresponsive to the body’s own molecules and cells, a breakdown that can initiate autoimmune disease. Normally, central tolerance removes or alters strongly self-reactive lymphocytes during their development, while peripheral tolerance restrains escaped cells through regulatory T cells, inhibitory signals, and limited access to target tissues; disruption of these controls can permit autoreactive T and B cells to activate and produce inflammatory responses or autoantibodies. Studying this process helps explain diseases such as type 1 diabetes, lupus, and rheumatoid arthritis, while informing biomarker development and therapies designed to restore immune regulation.
