Nadp+ Nadph

NADP+ and NADPH are interconvertible redox cofactors that transfer electrons in biological systems, supporting energy conversion, biosynthesis, and cellular protection. NADP+ accepts electrons and a hydrogen ion to form NADPH, while NADPH donates this reducing power to drive reactions that require electron input. In photosynthesis, light-dependent reactions generate NADPH, which fuels carbon fixation in the Calvin cycle; in cells, NADPH also supports fatty acid and nucleotide synthesis and helps maintain antioxidant defenses through systems such as glutathione. The NADP+/NADPH balance therefore links metabolism, redox regulation, and protection from oxidative stress.

NADPH oxidase is the major source of reactive oxygen species (ROS) in phagocytes. Because of the ephemeral nature of ROS, it is difficult to measure and monitor ROS levels in living animals. A minimally invasive method for serial quantification of ROS in living mice is described.

Determination of ROS Production by Neutrophils upon Interaction with Opsonized Biofilm

In this video, we demonstrate an in vitro chemiluminescence assay to detect the generation of reactive oxygen species by neutrophils upon exposure to an opsonized bacterial biofilm.

A Murine Skin Wound Infection Model to Study the Immune Response against a Bacterial Pathogen

This video demonstrates a technique to generate a murine model of skin wound infection. Upon infecting mice — engineered to express a fluorescent protein in neutrophils — with a genetically modified bioluminescent Staphylococcus aureus — a pathogenic bacteria. The bacterial burden at the infection site and the neutrophil recruitment dynamics can be studied using whole animal imaging.

Detecting Reactive Oxygen Species

2023

Reactive oxygen species are chemically active, oxygen-derived molecules capable of oxidizing other molecules. Because of their reactive nature, there are many deleterious effects associated with unchecked ROS production, including structural damage to DNA and other biological molecules. However, ROS can also be mediators of physiological signaling. There is accumulating evidence that ROS play significant roles in everything from activation of transcription factors to the mediation of...

Fluorescent Probe Imaging Assay: A Technique to Visualize Oxidative Stress in the Reactive Oxygen Species Inducer-Treated Cultured Intestinal Organoid Cells
