Proximal Tubule Transduction

Proximal tubule transduction is the introduction of genetic material into proximal tubule cells to modify or study gene expression in this specialized region of the kidney. Typically, a viral vector delivers DNA or RNA into cultured cells, where the introduced sequence can produce a target protein or alter cellular pathways involved in solute transport, metabolism, and injury responses. This method supports investigations of renal physiology, disease mechanisms, and nephrotoxicity by enabling controlled analysis of proximal tubule function. It can also help evaluate candidate genes, molecular treatments, and engineered kidney models while reducing reliance on whole-animal experiments.

Cellular ion transport can often be assessed by monitoring intracellular pH (pHi). Genetically Encoded pH-Indicators (GEpHIs) provide optical quantification of intracellular pH in intact cells. This protocol details the quantification of intracellular pH through cellular ex vivo live-imaging of Malpighian tubules of Drosophila melanogaster with pHerry, a pseudo-ratiometric genetically encoded pH-indicator.
