Resistance Fluctuation

Resistance fluctuation is the random variation in the number of resistant cells or colonies across independently grown microbial cultures, an observation central to understanding how heritable traits arise. During growth, spontaneous mutations can occur before exposure to an antibiotic or other selective agent; selection then allows resistant descendants to survive and reproduce, so cultures founded from similar cells may produce widely different resistance counts. Fluctuation analysis uses this uneven distribution to distinguish pre-existing genetic mutation from resistance induced by the environment. In biology, the approach clarifies antimicrobial resistance, supports quantitative studies of mutation rates, and explains how microbial populations can respond rapidly to treatment.

This video demonstrates the technique of fluorescence fluctuation spectroscopy (FFS) to study protein homo-oligomerization. Fluorescently-labeled proteins in a sample are dimerized using an agent when studying protein oligomerization using FFS. Using a confocal microscope, as the proteins move in and out of the small observation volume, the analysis of the fluctuations in the brightness of the fluorescent molecules is performed to determine the oligomeric state of the proteins.
