Spliceosome

The spliceosome is a dynamic molecular machine that removes noncoding introns from precursor messenger RNA and joins coding exons, enabling accurate gene expression in eukaryotic cells. It assembles on pre-mRNA from small nuclear ribonucleoproteins, or snRNPs, and associated proteins that recognize conserved splice sites and a branch-point sequence, then catalyze two sequential transesterification reactions to excise the intron and form mature mRNA. Spliceosome function influences transcript diversity through alternative splicing, allowing one gene to produce multiple RNA and protein variants. Studying its assembly and regulation helps explain development, cellular differentiation, and diseases caused by abnormal RNA processing.

Here, we describe the utilization of Grafix (Gradient Fixation), a glycerol gradient centrifugation in the presence of a crosslinker, to identify interactions between splicing factors that bind transiently to the spliceosome complex.

ACT1-CUP1 Assays Determine the Substrate-Specific Sensitivities of Spliceosomal Mutants in Budding Yeast

The ACT1-CUP1 assay, a copper growth assay, provides a quick readout of precursor messenger RNA (pre-mRNA) splicing and the impact mutant splicing factors have on spliceosomal function. This study provides a protocol and highlights the customization possible to address the splicing question of interest.

pre-mRNA Processing

2019

In eukaryotic cells, transcripts made by RNA polymerase are modified and processed before exiting the nucleus. Unprocessed RNA is called precursor mRNA or pre-mRNA to distinguish it from mature mRNA. Once about 20-40 ribonucleotides have been joined together by RNA polymerase, a group of enzymes adds a “cap” to the 5’ end of the growing transcript. In this process, a 5’ phosphate is replaced by modified guanosine that has a methyl group attached to it (7-Methyl guanosine). This 5’ cap helps the...

Alternative RNA Splicing

2020

Alternative RNA splicing is the regulated splicing of exons and introns to produce different mature mRNAs from a single pre-mRNA. Unlike in constitutive splicing where a single gene produces a single type of mRNA, alternative splicing allows an organism to produce multiple proteins from a single gene and plays an important role in protein diversity. There are five types of alternative RNA splicing that vary in the ways the pre-mRNA segments are removed or retained in the mature mRNA. The first...
