Tibial Growth Plate

The tibial growth plate is a layer of cartilage near the ends of the tibia that enables this long bone to grow in length during childhood and adolescence. Through endochondral ossification, resting chondrocytes develop into proliferating and hypertrophic cells arranged in distinct zones; their cartilage matrix is then replaced by mineralized tissue and bone on the shaft side of the plate. This coordinated process links cell division, matrix production, mineralization, and blood vessel invasion to skeletal growth. Studying the tibial growth plate helps explain limb development, growth disorders, and pediatric fractures, while providing a model for cartilage biology and bone repair.

The growth plate is a cartilaginous region in children's long bones where longitudinal growth occurs. When injured, bony tissue can form and impair growth. We describe a rat model of growth plate injury that leads to bony repair tissue, allowing the study of repair mechanisms and growth plate regeneration strategies.

Currently any kind of vascularized composite allotransplantation depends on long-term-immunosuppression, difficult to support for non-life-critical indications. We present a new porcine tibial VCA model that can be used to study bone VCA and demonstrate the use of surgical angiogenesis to maintain bone viability without the need of long-term immune-modulation.

Tricolor Transgenic Murine Model for Studying Growth Plate Injury

This protocol describes an improved mouse model for adolescent bone growth plate injuries. Using transgenic mice with tri-lineage fluorescent reporters for collagen types I, II, and X, the primary matrices associated with three different substrata of the growth plate, injury placement is guided by native fluorescence under the microscope.
