Iron Dependence

Iron dependence is the requirement for iron to support essential cellular functions, including oxygen transport, energy production, and DNA synthesis, and it is especially relevant to cancer biology. Cells obtain iron largely through transferrin uptake, store it in ferritin, and use it as a cofactor for enzymes involved in replication and mitochondrial respiration; excess free iron can also drive lipid peroxidation and ferroptosis. Many cancer cells increase iron acquisition to sustain rapid growth, creating potential vulnerabilities that researchers study through iron depletion, altered iron storage, and ferroptosis-based interventions. Understanding iron dependence may reveal biomarkers and guide therapies that selectively disrupt tumor metabolism while limiting damage to normal tissues.
