Organ Isolation

Organ isolation is the process of separating an intact organ or defined organ tissue from a donor organism or surrounding tissues for controlled study and analysis. It typically involves surgical dissection under sterile, carefully controlled conditions, followed by preservation of tissue architecture, viability, and relevant physiological properties during transport or ex vivo culture. In cancer research, isolated organs and organ fragments support the examination of tumor development, invasion, treatment response, and interactions between malignant cells and their tissue environment. These models can complement cell cultures and animal studies by retaining structural and cellular features that influence disease behavior and therapeutic outcomes.

We present protocols for the isolation of stable heterocyclic carbenes. The synthesis of a cyclic (alkyl)(amino) carbene (CAAC) and an N-heterocyclic carbene (NHC) is demonstrated using filter cannulas and Schlenk technique. We furthermore present the synthesis of the related oxygen-sensitive, electron-rich mixed “Wanzlick dimer” and the reduced stable organic radical.
