[4+2] Cycloaddition

A [4+2] cycloaddition is a pericyclic reaction in which a four-π-electron component, typically a conjugated diene, combines with a two-π-electron component, known as a dienophile, to form a six-membered ring. Usually proceeding through a concerted transition state under thermal conditions, the reaction reorganizes π bonds simultaneously and often preserves stereochemical relationships from the reactants. This transformation, commonly called the Diels–Alder reaction, can create two carbon–carbon bonds and multiple stereocenters in a single step. Chemists use it to construct cyclohexene frameworks efficiently in natural product synthesis, medicinal chemistry, and materials research.

Like benzene, cyclobutadiene and cyclooctatetraene are cyclic compounds with alternate single and double bonds. However, their chemical behavior differs from benzene, as they are unstable and not aromatic. So, what are the structural characteristics of unsaturated compounds categorized as aromatic? For the first time, Eric Hückel, a German chemical physicist, derived a set of structural features for a compound to be classified as aromatic. This is now known as Hückel’s rule or the 4n + 2 rule.
