Spectrophotometer Measurement

Spectrophotometer measurement is an analytical technique that determines how strongly a sample absorbs or transmits light at selected wavelengths, making it useful for identifying and quantifying chemical substances. A spectrophotometer passes light through a sample, compares the transmitted intensity with a reference, and calculates absorbance; under suitable conditions, the Beer–Lambert law relates absorbance to concentration and path length. In chemistry, this method supports concentration measurements, reaction monitoring, and characterization of compounds in solutions. Its speed, sensitivity, and relatively small sample requirements make it valuable in teaching laboratories, quality control, environmental analysis, and biochemical research.

The spectrophotometer is a routinely used instrument in scientific research. Spectrophotometry is the quantitative measurement of how much a chemical substance absorbs light by passing a beam of light through the sample using a spectrophotometer. In this video, basic concepts in spectrophotometry, including transmittance, absorbance and the Beer-Lambert Law are reviewed in addition to the components of the spectrophotometer. These concepts provide a foundation for how to determine the...

Polarimetry finds application in chemical kinetics to measure the concentration and reaction kinetics of optically active substances during a chemical reaction. Optically active substances have the capability of rotating the plane of polarization of linearly polarized light passing through them—a feature called optical rotation. Optical activity is attributed to the molecular structure of substances. Normal monochromatic light is unpolarized and possesses oscillations of the electrical field in...
