Thermo Responsive Polymers

Thermoresponsive polymers are materials that undergo predictable changes in solubility, swelling, conformation, or phase behavior when temperature changes, making them useful for controlling biological interactions. Their response commonly arises from a lower or upper critical solution temperature, at which the balance between polymer–water interactions and hydrophobic or hydrophilic forces shifts, causing reversible transitions such as chain collapse or expansion. In clinical research, these polymers support temperature-triggered drug delivery, injectable hydrogels, tissue engineering scaffolds, and cell-handling systems. Their tunable transition temperatures can enable localized release and minimally invasive administration, although biocompatibility, stability, and precise response control remain important design considerations.

We present a protocol to synthesize Janus microhydrogels composed entirely of the same base material, poly(N-isopropylacrylamide) (PNIPAAm), with a clearly compartmentalized structure base on the phase separation of a supersaturated NIPAAm monomer solution. The synthesized Janus microhydrogels show unique properties such as anisotropic thermo-responsiveness and organophilic/hydrophilic loading capability.

The word polymer is derived from the Greek words “poly” which means “many” and “mer” which means “parts”. Polymers are long chains of molecules composed of repeating units of smaller molecules, known as monomers. They either occur naturally, such as DNA and proteins, or can be constructed synthetically, like plastics. They have varied structural characteristics, such as linear chains, branched chains, or complex networks, that contribute to the properties that they exhibit. Additionally,...
