Decidualization Induction

Decidualization induction is the experimental initiation of decidualization, a hormone-responsive process in which uterine endometrial stromal cells differentiate to support embryo implantation and early pregnancy. In laboratory models, progesterone and cyclic adenosine monophosphate signaling, often combined with estrogen, activate transcriptional programs that alter cell morphology and stimulate secretion of markers such as prolactin and insulin-like growth factor-binding protein 1. Researchers use this approach to investigate endometrial receptivity, embryo implantation, maternal-fetal interactions, and disorders associated with impaired decidual responses. Controlled induction in primary cells or established cell models also provides a platform for comparing hormonal conditions and evaluating mechanisms of reproductive development.

A simple method to establish primary human placental (villous) and decidual organ cultures is described. Villous and decidual organ cultures are invaluable tools for studying pathogenesis at the human maternal-fetal interface. Infection with the facultative intracellular bacterium Listeria monocytogenes is demonstrated.
