Lugol's Iodine

Lugol's iodine is a laboratory solution of elemental iodine and potassium iodide used to detect and visualize polysaccharide-rich materials in biological samples. In solution, iodide increases iodine solubility and promotes polyiodide formation; these iodine species associate with carbohydrate structures, producing characteristic brown, blue, or blue-black staining depending on the material and its organization. In developmental biology, Lugol's iodine can help reveal glycogen, yolk-associated storage materials, and other carbohydrate-rich regions in embryos or tissues. This simple colorimetric reagent supports tissue characterization, developmental comparisons, and assessment of stored nutrients without requiring complex instrumentation.
