Syncytial Blastoderm

The syncytial blastoderm is an early embryonic stage in many insects, including Drosophila, in which nuclei divide within a shared cytoplasm before individual cell membranes form, making it a key model for studying development. During rapid, synchronous nuclear divisions, nuclei migrate toward the embryo’s cortex, where they eventually become enclosed by membranes during cellularization. Maternal factors and spatial signals regulate nuclear behavior and establish patterns of gene expression across the embryo. Research on the syncytial blastoderm has clarified how morphogen gradients, cell-cycle control, and early patterning coordinate embryonic development, while live imaging and genetic analysis continue to reveal the mechanisms governing these processes.

Generation of Naïve Blastoderm Explants from Zebrafish Embryos

Zebrafish blastoderm explants are generated by isolating embryonic cells from endogenous signaling centers within the early embryo, producing relatively naïve cell clusters easily manipulated and cultured ex vivo. This article provides instructions for making such explants and demonstrates their utility by interrogating roles for Nodal signaling during gastrulation.
