Negative Sequence

Negative sequence is a component of an unbalanced three-phase electrical system in which three equal-magnitude phasors are spaced 120° apart and rotate in the direction opposite to the positive-sequence component. Using symmetrical-component analysis, engineers decompose measured phase voltages or currents into positive-, negative-, and zero-sequence sets, allowing negative-sequence quantities to reveal phase imbalance and asymmetrical faults. Negative-sequence current produces a reverse-rotating magnetic field in generators and motors, which can cause rotor heating, torque pulsations, and reduced equipment life. Engineers therefore use negative-sequence measurements for fault detection, generator and motor protection, power-quality assessment, and diagnosing unbalanced operating conditions.

Generating Transposon Insertion Libraries in Gram-Negative Bacteria for High-Throughput Sequencing

We describe a method to generate saturating transposon mutant libraries in Gram-negative bacteria and subsequent preparation of DNA amplicon libraries for high-throughput sequencing. As an example, we focus on the ESKAPE pathogen, Acinetobacter baumannii, but this protocol is amenable to a wide range of Gram-negative organisms.
