Stress State Evaluation

Stress state evaluation is the engineering analysis of the internal forces acting at a point in a material, including their magnitude, direction, and distribution. It represents these loads with a stress tensor that resolves applied forces into normal and shear components on selected planes; coordinate transformations, principal stresses, and Mohr’s circle help identify critical orientations and combined loading effects. Engineers use this evaluation to assess strength, deformation, and failure risks in structures, machine components, and materials. The results support safer design, material selection, fatigue assessment, and interpretation of experimental or computational stress data under complex loading conditions.

Induction and Evaluation of Depression in Mice Through Chronic Stress Exposure

2025

Source: Yan, Z., et al. Evaluating the Anti-depression Effect of Xiaoyaosan on Chronically-stressed Mice. J. Vis. Exp. (2019).The video demonstrates the process of establishing a mouse model of chronic stress-induced depression and assessing depressive behaviors. The chronic application of various stressors triggers the release of corticosterone, leading to neuronal over-excitation and eventual neuronal death, which results in depression-like symptoms in the mice.

This article describes a protocol for inducing psychological stress in participants, which enables researchers to measure psychological, physiological and neuroendocrine responses to stress within single participants or between groups.

Evaluating the Effect of Beneficial Bacteria on Gut Microbial Translocation in Heat-Stressed Rats

Source: Sorokulova, I., et al. Prevention of Heat Stress Adverse Effects in Rats by Bacillus subtilis Strain. J. Vis. Exp. (2016)This video demonstrates rats pre-treated with beneficial bacteria or phosphate-buffered saline (PBS) and exposed to heat stress to evaluate gut barrier integrity. Fewer gut-derived bacterial colonies from the liver of treated rats, in contrast to numerous colonies in control rats, indicate an intact epithelial barrier and minimal bacterial translocation.

A Spot Plate Assay for Evaluating Oxidative Stress Responses in Bacterial Strains

2025

This video demonstrates a spot plate assay tailored to assess the oxidative stress responses of bacterial strains. Some strains produce pyomelanin, an antioxidant pigment. Conversely, strains treated with a test agent, leading to reduced pyomelanin production, exhibit increased susceptibility to oxidative stress, resulting in decreased cell viability compared to pyomelanin-producing strains.
