4-hau Antigen

4-HAU antigen is a standardized quantity of hemagglutinating antigen used to measure antibodies that inhibit viral hemagglutination in immunology and infection research. The antigen is diluted until it produces four hemagglutination units, reflecting its ability to cross-link red blood cells through interactions between viral surface proteins and receptors on the cells. In a hemagglutination-inhibition assay, patient serum or experimental antisera are mixed with the 4-HAU preparation before red blood cells are added; inhibition indicates antigen-specific antibodies. This defined antigen dose improves assay consistency and supports serological diagnosis, immune-response measurement, vaccine evaluation, and comparison of antibody titers across samples.

Here, we present an overview of the preparation and animal handling procedures required to safely perform medical imaging in an animal biosafety level 4 laboratory. Computed tomography of a mock-infected guinea pig illustrates these procedures that may be used to evaluate the disease caused by a high consequence pathogen.
