Cryopreserved Monocytes

Cryopreserved monocytes are blood-derived innate immune cells stored at very low temperatures to preserve them for later research, enabling standardized studies of inflammation and host responses to infection. Before freezing, monocytes are suspended with a cryoprotectant such as dimethyl sulfoxide and cooled under controlled conditions, then maintained in liquid nitrogen; rapid, careful thawing limits ice-related cell injury and supports recovery. After thawing, they can be assessed directly or differentiated into macrophages or dendritic cells using defined culture signals. These cells provide a practical, reproducible model for examining pathogen recognition, cytokine production, antimicrobial responses, immune activation, and therapeutic effects while reducing dependence on same-day fresh blood samples.

Culture of Macrophage Colony-stimulating Factor Differentiated Human Monocyte-derived Macrophages

A protocol is presented for cell culture of macrophage colony-stimulating factor (M-CSF) differentiated human monocyte-derived macrophages. The protocol utilizes cryopreservation of monocytes coupled with their bulk differentiation into macrophages. Then harvested macrophages can then be seeded into culture wells at required cell densities for carrying out experiments.

Mouse Sperm Cryopreservation and Recovery using the I·Cryo Kit

Here we demonstrate the newly developed I•Cryo kit for mouse sperm cryopreservation. Two-cell stage embryo development with frozen-thawed sperm was improved consistently in 5 mouse strains with the use of this kit. Over a 1.5 year period, 49 genetically modified mouse lines were archived by sperm cryopreservation with the I•Cryo kit and later successfully recovered by IVF.

The isolation of neonatal rat cardiomyocytes is a time consuming and unpredictable procedure. This study describes methods for cryopreservation and thawing of neonatal rat cardiomyocytes that allows for more efficient use of cells. The thawed NRCMs can be used for various experiments without the need for performing isolations each time.

This study compares two different methods of human monocyte isolation for obtaining in vitro dendritic cells (DCs). Monocytes are selected by adherence or negatively enriched by magnetic separation. Monocyte yield and viability along with MDDC viability, proliferation and CD11c/CD14 surface marker expression will be compared between both methods.

Cryopreservation of Preimplantation Embryos of Cattle, Sheep, and Goats

Preimplantation embryos may be cryopreserved after placement into a hypertonic cryoprotective solution to cause cellular dehydration. After equilibration, ice crystal formation is induced in the solution surrounding the embryo. Further dehydration occurs as the embryo is slowly cooled to subzero temperatures before plunging into liquid nitrogen for storage.
