Murine Leukemia Virus Core

The Murine Leukemia Virus (MLV) core is the virus’s internal protein-nucleic acid complex, enclosed by the viral envelope and essential for protecting and delivering the retroviral genome. It contains the RNA genome associated with nucleocapsid proteins, together with viral enzymes such as reverse transcriptase and integrase; after the virus enters a host cell, core disassembly releases these components so the RNA can be copied into DNA and integrated into the host genome. Studying the MLV core clarifies retroviral assembly, entry, uncoating, and replication, while supporting research on viral vectors, host immune recognition, and mechanisms of infection.

This manuscript provides a technical procedure that can be used to characterize C1498 cell cultures in vitro and the acute leukemia induced in mice after their injection. Phenotypic and functional analyses are performed using flow cytometry, immunofluorescence microscopy, cytochemistry and May-Grünwald Giemsa staining.
