Mutagen Resistance Screening

Mutagen resistance screening is a set of assays used to determine how cells or microorganisms withstand DNA-damaging chemical or physical agents, an important question in genetics, infection biology, and toxicology. Experiments typically expose cultures to defined mutagen concentrations, then measure survival, growth recovery, or mutation-associated phenotypes against untreated controls; differences can reveal protective barriers or DNA repair capacity. In immunology and infection research, these screens help characterize pathogen stress tolerance, identify DNA repair pathways that influence persistence, and compare candidate antimicrobial or genotoxic interventions while supporting risk assessment and mechanistic studies of mutation and adaptation.

Mutagenicity and Carcinogenicity

2023

Mutagenicity and carcinogenicity refer to the ability of drugs to cause genetic defects and induce cancer, respectively. The International Agency for Research on Cancer (IARC) classifies agents into four groups based on their carcinogenic potential. Group 1 agents are known human carcinogens; group 2A agents are probably carcinogenic to humans; group 3 agents lack data to support their role in carcinogenesis; and group 4 includes agents for which data support that they are not likely to be...

Source: Hassan, J. A., et al. High-Throughput Identification of Resistance to Pseudomonas syringae pv. Tomato in Tomato using Seedling Flood Assay. J. Vis. Exp. (2020).This video demonstrates a method to screen for bacterial resistance in different tomato strains using ten-day-old seedlings inoculated with a bacterial suspension. It explains how bacteria invade through stomata, deliver effector proteins to suppress plant immunity, and how plant responses differ between susceptible and resistant...

Forward Genetic Approach to Uncover Stress Resistance Genes in Mice — A High-throughput Screen in ES Cells

2015

Stress resistance is one of the hallmarks for longevity and is known to be genetically governed. Here, we developed an unbiased high-throughput method to screen for mutations that confer stress resistance in ES cells with which to develop mouse models for longevity studies.

Virus-induced gene silencing is an useful tool for identifying genes involved in nonhost resistance of plants. We demonstrate the use of bacterial pathogens expressing GFPuv in identifying gene silenced plants susceptible to nonhost pathogens. This approach is easy, fast and facilitates large scale screening and similar protocol can be applied to studying various other plant-microbe interactions.
